Job description
Transcriptional regulation is the molecular basis that drives general cellular processes and cell-type specification in multicellular organisms. The multi-protein BAF chromatin remodeling complex is a transcriptional regulator that is recruited by pioneer transcription factors (TFs) and assists the latter to control gene expression in mammalian cells. Due to functional importance of this complex, mutations of the components in this complex are found in many types of diseases including neurodevelopmental disorders and >20% of human malignancies. This project aims to understand the functions of different BAF complex subunits in gene regulation.
We are looking for a highly motivated PhD student who has strong interests and dedication to basic gene regulation research. In the first phase of this project, we will use pluripotent stem cells to understand the effects of different BAF mutations on chromatin and transcription. In the second phase, we will further differentiate pluripotent stem cells into other 2D or 3D culture, such as neural progenitors and bone marrow organoids, to understand cell type specific effects of the BAF mutations.
Work environment
You will be a part of small research team in the department of hematology at the Erasmus Medical Center in Rotterdam. At this moment, the lab is consisted of are 1 PhD student and 1 research technician. Also, we closely work with other research groups with similar interests at the Erasmus MC and in other Dutch institutes.
Qualifications and skills
Requirements:
1. An MSc in either (molecular) biology, genetics, and/or (bio)chemistry;
2. Solid experience with molecular biology techniques (e.g. molecular cloning, gene editing) and mammalian tissue culture (e.g. 2D/3D culture);
3. A strong motivation for performing gene regulation research;
4. Proficient in written and spoken English.
Desirable skills:
1. You have performed internship in a gene regulation lab (e.g. chromatin biology, transcription, 3D genome);
2. You are familiar with biochemistry techniques such as immunoprecipitation;
3. You have experience with analyzing genomic data.

Terms of employment
Fixed-term contract: 4 years.
You will receive a temporary position for 4 years. The gross monthly salary is ‚ € 3.017,- in the 1st year and gradually increases to‚ € 3.824,- in the 4th year (scale OIO).
Excellent fringe benefits, such as a 13th month that is already paid out in November and an individual travel expense package.
An International Office which aids you in preparing for you arrival and stay.
Pension insurance with ABP. We take care of approximately 2/3 of the monthly contribution.
